Purification
DNA purification starts as soon as a donor closes the collection vial and the saliva is mixed with the Oragene•DNA solution. While the purifier is included with the Oragene•DNA kits, some researchers and clinicians prefer to continue extracting the DNA using their existing protocols. Oragene•DNA is fully compatible with other manual extraction protocols and automated robots for high throughput customers.
The documents in this section will provide you with additional information about purifiying DNA from Oragene•DNA/saliva samples.
Manual Purification Protocols
Laboratory Protocol for Manual Purification of DNA from 500 µL
Sample of Oragene®•DNA using the Agencourt DNAdvance Kit (pdf):
This protocol details the purification process using Agencourt's DNAdvance Kit for saliva collected using Oragene•DNA Self-Collection Kits.
Laboratory Protocol for Manual Purification of DNA from 0.5 mL of Oragene•DNA / saliva (pdf):
Step-by-step instructions for extracting DNA from 0.5 mL of Oragene•DNA/saliva samples.
Laboratory Protocol for Manual Purification of DNA from 4.0 mL of Oragene•DNA / saliva (pdf):
Step-by-step instructions for extracting DNA from 4.0 mL of Oragene•DNA/saliva samples.
Oragene•DNA purification using the PUREGENE® DNA Purification Kit (pdf):
Purification protocol for extracting DNA from an Oragene•DNA/saliva sample using the Puregene DNA Purification Kit (Gentra).
Compatibility of the PUREGENE® DNA Purification Kit with Oragene•DNA Collection (pdf):
DNA samples collected with Oragene•DNA and purified with Puregene have an average DNA yield of 85.5 µg and an A260/A280 ratio of 1.79. The DNA is high-molecular weight and suitable for downstream applications like PCR.
Comparison of DNA purified with Oragene•DNA and the QIAamp Mini Kit (pdf):
Saliva samples collected with Oragene•DNA may be purified using the Oragene•DNA purification protocol or the QIAamp DNA Mini Kit (Qiagen).
Compatibility with Automated Purification Systems
Automated extraction of gDNA from Oragene•DNA / saliva samples
using the Tecan Freedom EVO liquid handling platform and the
Promega MagaZorb DNA Mini-Prep kit (pdf):
OrageneDNA facilitates the non-supervised, noninvasive
collection of gDNA from a large number
of individuals. In order to assist in the processing of
such large numbers of samples we have validated the
purifi cation of OrageneDNA samples using the Tecan
Freedom EVO liquid handling robot in conjunction with
the Promega MagaZorb DNA Mini-Prep kit.
Compatibility of the AUTOPURE® with Oragene•DNA DNA Collection (pdf):
DNA samples collected with Oragene•DNA and purified with the Autopure LS instrument have an average DNA yield of 82.3 µg and an A260/A280 ratio of 1.87. The DNA is high-molecular weight and suitable for downstream applications like PCR.
High-throughput DNA purification with the Qiagen BioRobot EZ1 (pdf):
The BioRobot EZ1 is an automated high-throughput DNA purification system that uses silica-magnetic-particle technology. The EZ1 can purify up to six Oragene•DNA/saliva samples in less than 20 minutes, with excellent DNA recovery.
High-throughput DNA purification with the Magtration® 12GC (pdf):
The Magtration 12GC is an automated high-throughput DNA purification system that uses paramagnetic-particle technology. The 12GC can purify up to twelve Oragene•DNA/saliva samples in 30 minutes, with excellent DNA yields.
Purification Protocol for 4ml of Oragene Saliva (Chemagen) (pdf)
This protocol details the purification process using the chemagic Magnetic Separation Module I and the chemagic Dispenser for saliva collected using Oragene•DNA Self-Collection Kits.
